Emergency services were called to the scene of a collision in the village of Duagh near Listowel at about 10 to 10 last night.
A pedestrian, aged in his late teens, was seriously injured, and later passed away.
The driver, a woman in her early 20s, and three passengers, all in their late teens, were uninjured.
Another pedestrian, in his 40s, was hit by a car on the M1 motorway at Junction 16 southbound near Dundalk at about 2.45 this morning.
The man in his 50s was driving and his three passengers were treated at the scene for non life-threatening injuries.
Separately, a man in his early 20s was killed at about 2.50 morning when the car he was driving overturned on Scrine Hill near Athleague in County Roscommon.
Gardaí remain at the scene of the three collisions today, as technical examinations of the scenes get underway, and post-mortems will be carried out in due course.
